Research summary
Human studies in healthy volunteers show that alvimopan can reverse opioid-induced delays in gastrointestinal transit and accelerate colonic transit, while not significantly affecting centrally mediated opioid effects such as analgesia or pupillary constriction. Pharmacokinetic studies in healthy subjects also show measurable oral absorption and predictable exposure across dose ranges studied. Overall, the evidence base in healthy humans supports a clear physiologic effect on opioid-slowed gut motility, but not broad health or performance benefits. The strongest clinical evidence for therapeutic benefit comes from surgical populations rather than healthy adults, so for healthy people this remains primarily a mechanistic research compound rather than a general supplement.
