Research summary

Human research on ambenonium chloride focuses almost entirely on its use as a therapeutic drug in patients with myasthenia gravis, examining clinical efficacy, pharmacokinetics, organ selectivity, and safety. Studies in myasthenic patients demonstrate that orally administered ambenonium can improve neuromuscular transmission and muscle strength, but also reveal substantial interindividual variability in serum levels and an erratic relationship between dose and exposure, which complicates dose optimization and disease control. In vitro work using human erythrocyte acetylcholinesterase and related assays confirms that ambenonium is a highly potent, reversible inhibitor with mixed competitive and uncompetitive characteristics, helping to explain its rapid onset of action and strong cholinergic effects. However, there are no controlled studies of ambenonium in healthy human volunteers for performance, cognition, or general wellness, and the compound is not approved or studied as a dietary supplement.
