Amlodipine

Randomized trial comparing amlodipine 5-10mg to nifedipine GITS in 500 hypertensive patients to assess effect on morning blood pressure surge via 24-hour ambulatory monitoring.

In 635 patients undergoing percutaneous transluminal coronary angioplasty, pre-treatment with amlodipine 10mg reduced the requirement for repeat PTCA compared to placebo (3.1% vs 7.3%).

In normotensive patients treated with amlodipine, there was a decline in cardiovascular events, evidence of regression of atherosclerotic changes, fewer hospitalizations, and significant reductions in non-fatal myocardial infarction by 26% and stroke or transient ischaemic attack by 50%.

In 19,342 hypertensive patients, amlodipine-based regimen (with perindopril as required) reduced non-fatal myocardial infarction, fatal coronary heart disease, stroke, total cardiovascular events, all-cause mortality, and incident diabetes compared to atenolol-based treatment.

In over 33,000 hypertensive patients with at least one coronary heart disease risk factor, amlodipine as a calcium channel blocker was compared to diuretics and ACE inhibitors, showing comparable outcomes in preventing coronary heart disease and other cardiovascular events.
