Glyburide

A placebo-controlled randomized trial in 21 healthy, non-diabetic volunteers receiving 1.5 mg daily glyburide for 7 days while under inpatient monitoring. The study evaluated safety through neuropsychological testing, physical exams, vital signs, ECGs, cognitive assessments, and frequent serum glucose measurements to assess for adverse events including hypoglycemia and cognitive impacts.

A study in 9 healthy subjects examining the relationship between plasma glyburide concentrations and insulin response during euglycemic and hyperglycemic conditions. Results showed that glyburide enhanced insulin secretion and glucose disposal only at drug levels of 100-200 nM, corresponding to oral doses of 10 mg or less, suggesting a narrow therapeutic window.

Blood glucose levels were significantly lowered by glyburide and both metabolites through increased insulin secretion, with oral glyburide showing the greatest effect (23.8% AUC reduction).
