Meglitinides
medication Under reviewMeglitinides are a class of prescription oral diabetes medications, including repaglinide and nateglinide. They are taken shortly before meals and work by stimulating the pancreas to release insulin, which helps reduce after-meal blood sugar spikes. They are used mainly to improve glucose control in type 2 diabetes, especially postprandial glucose. Compared with longer-acting insulin secretagogues, they are shorter acting and are generally associated with a lower risk of prolonged hypoglycemia, though low blood sugar and weight gain can still occur.
Research summary
The clinical evidence in humans shows that meglitinides lower HbA1c and post-meal glucose in people with type 2 diabetes, with effects generally comparable to other oral glucose-lowering drugs in some trials and reviews. However, the evidence base does not address benefits in healthy subjects because these drugs are studied as treatments for diabetes rather than as supplements or performance enhancers. Among human studies, the main documented adverse effects are hypoglycemia, weight gain, and occasional gastrointestinal or neurologic symptoms. Drug-interaction literature also shows that some CYP inhibitors can raise repaglinide exposure, increasing hypoglycemia risk.
