Research summary
Clinical research on pyrimethamine in healthy human subjects is limited. Available studies demonstrate that intravenous pyrimethamine is well-tolerated in healthy volunteers with a half-life of approximately 140 hours, showing no significant changes in vital signs, electrocardiogram, hematology, or biochemical parameters. Oral administration studies in patients with late-onset GM2 gangliosidosis found that doses of 50 mg per day or lower were well-tolerated and produced therapeutic enzyme enhancement, while doses at or above 75 mg per day were associated with significant adverse effects including gastrointestinal symptoms and neurological complications. The pharmacokinetics of pyrimethamine show marked individual variation among subjects.
