Research summary
The human evidence base for tolbutamide in healthy subjects is limited and mostly mechanistic. Studies in normal volunteers show that tolbutamide can rapidly stimulate insulin secretion and produce hypoglycemia, and that the magnitude of these effects varies substantially between individuals, partly because of differences in drug metabolism. Research in healthy adults also shows that tolbutamide metabolism is genetically variable and that its effects can be influenced by interactions with other drugs that share metabolic pathways. Overall, tolbutamide is not supported as a health supplement for healthy people. The human literature mainly supports its use as a glucose-lowering pharmacologic agent and as an experimental probe for insulin and glucagon physiology rather than for broad health benefits.
